Il s\u2019agit d\u2019une maladie chronique qui affecte les art\u00e8res et progresse par la formation de \u00abplaques d\u2019ath\u00e9rome\u00bb, lesquelles r\u00e9sultent de la r\u00e9tention et de l\u2019oxydation de lipoprot\u00e9ines de faible densit\u00e9, appel\u00e9es LDL (<em>Low Density Lipoproteins<\/em>), et d&rsquo;une inflammation incontr\u00f4l\u00e9e. Cette inflammation conduit \u00e0 la rupture des plaques, provoquant ainsi des accidents cardiovasculaires.<\/p>\n<p>Les macrophages jouent un r\u00f4le \u00e0 toutes les \u00e9tapes du d\u00e9veloppement des plaques d&rsquo;ath\u00e9rome. Certains macrophages sont ath\u00e9rog\u00e8nes, comme les macrophages inflammatoires et les macrophages spumeux \u00e0 un stade avanc\u00e9, tandis que d\u2019autres pourraient avoir un r\u00f4le ath\u00e9roprotecteur, tels que les macrophages immunor\u00e9gulateurs.<\/p>\n<p>Le <strong>premier objectif<\/strong> de nos projets est de d\u00e9velopper des outils permettant de discriminer les diff\u00e9rentes sous-populations de macrophages tout au long de l\u2019arbre art\u00e9riel, afin d\u2019identifier les plaques vuln\u00e9rables. Deux strat\u00e9gies sont \u00e9tudi\u00e9s&nbsp;:<\/p>\n<ol>\n<li>Exploiter l\u2019autofluorescence naturelle des macrophages li\u00e9e \u00e0 leur m\u00e9tabolisme (Labex TRAIL MEMIM \/ ANR MEAP\/ FFC COMEMAPA)<\/li>\n<li>Identifier des anticorps sp\u00e9cifiques des diff\u00e9rents types de macrophages (pro-ath\u00e9rog\u00e8nes versus antiath\u00e9rog\u00e8nes) (FFC PolAtheraDiag, ANR MSREI, EIC Pathfinder Open Horizon ABCardionostics)<\/li>\n<li>Utiliser des nanoparticules ou mol\u00e9cules biomim\u00e9tiques ciblant les macrophages spumeux (ANR NanoBioID, ANR LPCBioMab)<\/li>\n<\/ol>\n<p>Le <strong>deuxi\u00e8me objectif<\/strong> explore plusieurs pistes de th\u00e9rapies ciblant les macrophages (ANR NanoBioID, ANR LPCBioMab,&nbsp; EIC Pathfinder Open Horizon ABCardionostics) :<\/p>\n<ol>\n<li>Bloquer la polarisation des macrophages vers l\u2019\u00e9tat inflammatoire et\/ou induire un changement ph\u00e9notypique et fonctionnel vers un \u00e9tat immunor\u00e9gulateur<\/li>\n<li>Restaurer les fonctions ath\u00e9roprotectrices, telles que l\u2019efflux du cholest\u00e9rol, en optimisant ou en ciblant l\u2019apport d\u2019apoprot\u00e9ine ApoA1* au niveau des plaques.<\/li>\n<\/ol>\n<p>*<em>L\u2019ApoA1 est pr\u00e9sente dans les mol\u00e9cules de HDL, qui, dans des conditions normales, induisent l\u2019efflux du cholest\u00e9rol. Il s\u2019agit d\u2019une maladie chronique qui affecte les art\u00e8res et progresse par la formation de \u00abplaques d\u2019ath\u00e9rome\u00bb, lesquelles r\u00e9sultent de la r\u00e9tention et de l\u2019oxydation de lipoprot\u00e9ines de faible densit\u00e9, appel\u00e9es LDL (Low Density Lipoproteins), et d&rsquo;une inflammation incontr\u00f4l\u00e9e. Cette inflammation conduit \u00e0 la rupture des plaques, provoquant ainsi des accidents cardiovasculaires. Les macrophages jouent un r\u00f4le \u00e0 toutes les \u00e9tapes du d\u00e9veloppement des plaques d&rsquo;ath\u00e9rome. Certains macrophages sont ath\u00e9rog\u00e8nes, comme les macrophages inflammatoires et les macrophages spumeux \u00e0 un stade avanc\u00e9, tandis que d\u2019autres pourraient avoir un r\u00f4le ath\u00e9roprotecteur, tels que les macrophages immunor\u00e9gulateurs. Le premier objectif de nos projets est de d\u00e9velopper des outils permettant de discriminer les diff\u00e9rentes sous-populations de macrophages tout au long de l\u2019arbre art\u00e9riel, afin d\u2019identifier les plaques vuln\u00e9rables. Deux strat\u00e9gies sont \u00e9tudi\u00e9s&nbsp;: Exploiter l\u2019autofluorescence naturelle des macrophages li\u00e9e \u00e0 leur m\u00e9tabolisme (Labex TRAIL MEMIM \/ ANR MEAP\/ FFC COMEMAPA) Identifier des anticorps sp\u00e9cifiques des diff\u00e9rents types de macrophages (pro-ath\u00e9rog\u00e8nes versus antiath\u00e9rog\u00e8nes) (FFC PolAtheraDiag, ANR MSREI, EIC Pathfinder Open Horizon ABCardionostics) Utiliser des nanoparticules ou mol\u00e9cules biomim\u00e9tiques ciblant les macrophages spumeux (ANR NanoBioID, ANR LPCBioMab) Le deuxi\u00e8me objectif explore plusieurs pistes de th\u00e9rapies ciblant les macrophages (ANR NanoBioID, ANR LPCBioMab,&nbsp; EIC Pathfinder Open Horizon ABCardionostics) : Bloquer la polarisation des macrophages vers l\u2019\u00e9tat inflammatoire et\/ou induire un changement ph\u00e9notypique et fonctionnel vers un \u00e9tat immunor\u00e9gulateur Restaurer les fonctions ath\u00e9roprotectrices, telles que l\u2019efflux du cholest\u00e9rol, en optimisant ou en ciblant l\u2019apport d\u2019apoprot\u00e9ine ApoA1* au niveau des plaques. *L\u2019ApoA1 est pr\u00e9sente dans les mol\u00e9cules de HDL, qui, dans des conditions normales, induisent l\u2019efflux du cholest\u00e9rol. Cependant, elle est souvent oxyd\u00e9e et devient inefficace dans les conditions pathologiques.
