data-end=\"247\">Neuroenergetics<\/strong> \u2014 how the brain generates energy to sustain its functions \u2014 is gaining growing interest, from enhancing cognitive performance through lifestyle factors like exercise and nutrition, to developing neuroprotective strategies against neurodegenerative diseases. Understanding the cellular mechanisms linking neuronal activity and energy homeostasis is key to uncovering how neurons meet their high energy demands.<\/p><p class=\"\" data-start=\"659\" data-end=\"1037\">\ud83d\udd2c Our team has long investigated the <strong data-start=\"697\" data-end=\"744\">Astrocyte-Neuron Lactate Shuttle Hypothesis<\/strong> (ANLSH,\u00a0<em><a href=\"https:\/\/www.pnas.org\/doi\/epdf\/10.1073\/pnas.91.22.10625\">Pellerin and Magistretti, PNAS 1994<\/a><\/em>). Using NMR spectroscopy and, more recently, <strong data-start=\"834\" data-end=\"845\">in vivo<\/strong> functional MRI and MRS in genetically modified rats, we are studying whether lactate transfer occurs in the living brain and its importance for neuronal function (<em><a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC8617497\/pdf\/pnas.202112466.pdf\">Roumes et al. PNAS 2021<\/a><\/em>).<\/p><p class=\"\" data-start=\"1039\" data-end=\"1147\">\ud83d\udca1 This raises a central question: if lactate is a good neuronal fuel, could it also be <strong data-start=\"1127\" data-end=\"1146\">neuroprotective<\/strong>?<\/p><p class=\"\" data-start=\"1149\" data-end=\"1441\">\ud83d\udc00 Building on our discoveries, we are exploring therapeutic strategies for <strong data-start=\"1225\" data-end=\"1254\">neonatal hypoxia-ischemia<\/strong>\u2014a leading cause of infant mortality and disability, with no current pharmacological treatment. We showed that <strong data-start=\"1365\" data-end=\"1410\">lactate administration is neuroprotective<\/strong> (<em><a href=\"https:\/\/www.rmsb.u-bordeaux.fr\/wp-content\/uploads\/2020\/04\/2020JCBFM_ROUMES.pdf\">Roumes et al., JCBFM 2021<\/a><\/em>).\u00a0<span style=\"font-family: var( --e-global-typography-text-font-family ), Sans-serif; text-align: var(--text-align); caret-color: #000000; color: #000000; font-size: medium;\">\ud83e\uddec <span style=\"color: #808080;\">Based on our fondamental discoveries , we are also developing new projects aimed at <\/span><\/span><span style=\"color: #808080;\"><strong style=\"font-family: var( --e-global-typography-text-font-family ), Sans-serif; text-align: var(--text-align); font-size: 1rem; caret-color: #000000;\" data-start=\"311\" data-end=\"374\">modulating brain metabolism to counteract neurodegeneration<\/strong><span style=\"font-family: var( --e-global-typography-text-font-family ), Sans-serif; text-align: var(--text-align); caret-color: #000000; font-size: medium;\">. Because early metabolic dysfunction is increasingly recognized as a driver of neuronal vulnerability, we are investigating how targeting key metabolic regulators\u2014such as the astrocyte\u2013neuron lactate shuttle, alternative energetic substrates, or hormone-driven metabolic pathways\u2014might restore energy homeostasis and provide neuroprotection. <strong>These studies open new translational perspectives for metabolic-based interventions in neurodegenerative diseases<\/strong>.<\/span><\/span><\/p><p class=\"\" data-start=\"1443\" data-end=\"1738\">\ud83c\udf47 We also study how <strong data-start=\"1464\" data-end=\"1502\">nutrition affects brain metabolism<\/strong>. We&rsquo;ve demonstrated that <strong data-start=\"1528\" data-end=\"1543\">polyphenols<\/strong>, beyond their anti-inflammatory and antioxidant effects, modulate energy metabolism and enhance the expression of genes and proteins involved in the ANLS (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7844160\/pdf\/fnins-14-616824.pdf\"><em>Dumont et al., Front in Neurosci 2020<\/em><\/a>).<\/p><p class=\"\" data-start=\"1740\" data-end=\"2032\">\ud83d\udc76 Our main focus is on <strong data-start=\"1764\" data-end=\"1795\">neuroenergetics in neonates<\/strong>, which differs from adults and remains poorly understood. performance through lifestyle factors like exercise and nutrition, to developing neuroprotective strategies against neurodegenerative diseases. Understanding the cellular mechanisms linking neuronal activity and energy homeostasis is key to uncovering how neurons meet their high energy demands. \ud83d\udd2c Our team has long investigated the Astrocyte-Neuron Lactate Shuttle Hypothesis (ANLSH,\u00a0Pellerin and Magistretti, PNAS 1994). Using NMR spectroscopy and, more recently, in vivo functional MRI and MRS in genetically modified rats, we are studying whether lactate transfer occurs in the living brain and its importance for neuronal function (Roumes et al. PNAS 2021). \ud83d\udca1 This raises a central question: if lactate is a good neuronal fuel, could it also be neuroprotective? \ud83d\udc00 Building on our discoveries, we are exploring therapeutic strategies for neonatal hypoxia-ischemia\u2014a leading cause of infant mortality and disability, with no current pharmacological treatment. We showed that lactate administration is neuroprotective (Roumes et al., JCBFM 2021).\u00a0\ud83e\uddec Based on our fondamental discoveries , we are also developing new projects aimed at modulating brain metabolism to counteract neurodegeneration. Because early metabolic dysfunction is increasingly recognized as a driver of neuronal vulnerability, we are investigating how targeting key metabolic regulators\u2014such as the astrocyte\u2013neuron lactate shuttle, alternative energetic substrates, or hormone-driven metabolic pathways\u2014might restore energy homeostasis and provide neuroprotection. These studies open new translational perspectives for metabolic-based interventions in neurodegenerative diseases. \ud83c\udf47 We also study how nutrition affects brain metabolism. We&rsquo;ve demonstrated that polyphenols, beyond their anti-inflammatory and antioxidant effects, modulate energy metabolism and enhance the expression of genes and proteins involved in the ANLS (Dumont et al., Front in Neurosci 2020). \ud83d\udc76 Our main focus is on neuroenergetics in neonates, which differs from adults and remains poorly understood.
